A study investigated the link between thyroid hormone levels and osteoarthritis (OA), examining how triiodothyronine (T3) affects hypertrophic chondrocyte differentiation and OA progression. The study used two-sample Mendelian randomization and found a positive causal association between hyperthyroidism and OA. Analysis of RNA-sequencing data identified genes influenced by T3, with validation showing increased expression of MAFB, C1QTNF1, COL3A1, and ANGPTL2. These genes demonstrated strong diagnostic potential for OA. The findings suggest hyperthyroidism promotes OA progression via T3-induced chondrocyte changes, highlighting these genes as potential diagnostic markers.
