T-cell dysfunction in natural killer/T-cell lymphoma

Natural killer/T-cell lymphoma (NKTCL) is an aggressive T-cell lymphoma associated with Epstein‒Barr virus (EBV) infection. This study examined T-cell dysfunction in NKTCL patients and its impact on the immune microenvironment. The researchers found that NKTCL patients had higher frequencies of inhibitory T regulatory cells (Tregs) and myeloid-derived suppressor cells (MDSCs) compared to healthy donors. T cells from NKTCL patients exhibited higher expression of inhibitory receptors (IRs) and reduced proliferation and cytokine production. EBV-specific cytotoxic cells were decreased in NKTCL patients and showed upregulation of IRs. NKTCL cells induced T-cell exhaustion phenotypes and the generation of Tregs and MDSCs. These findings highlight the immune dysregulation in NKTCL and its potential impact on antitumor immunity.
