The present study discusses the expanding literature on the gasdermin (GSDM) family of proteins and their role in regulating inflammatory cell death beyond pyroptosis. The article explicitly highlights the critical contributions of GSDMs in the gastrointestinal system and their involvement in gastrointestinal cancers, enteric infections, and gut mucosal inflammation. The article also addresses several controversial issues surrounding GSDM biology, including their lytic versus non-lytic functions, the role of cleaved versus full-length proteins, and whether they promote pathogenic or protective effects in specific disease settings. Finally, the article summarizes the current literature on GSDM biology and highlights future research areas to unravel their particular role(s).
