The COVID-19 pandemic has reversed the progress made in global tuberculosis (TB) incidence and mortality in 2021, highlighting the need for investments in improving TB treatment outcomes. Recent randomized controlled trials have shown that a 4-month regimen of isoniazid, rifapentine, pyrazinamide, and moxifloxacin is non-inferior to the standard 6-month regimen for drug-susceptible pulmonary TB. New regimen backbones, such as bedaquiline-rifamycin and pretomanid-rifamycin, are also being evaluated for shorter treatment periods. In this context, Dooley and colleagues have evaluated pretomanid-rifamycin combinations for drug-susceptible TB, substituting pretomanid for ethambutol.
