UBTF Tandem Duplications Offer Clues for Improved AML Diagnosis and Treatment

A recent study found that tandem duplications (TDs) of the UBTF gene occur in about 3% of adult patients aged 18-60 years with acute myeloid leukemia (AML). UBTF-TDs were found to be mutually exclusive with other known AML subtype-defining alterations. The characteristics of UBTF-TD AML included young age, low bone marrow blast infiltration, and high rates of WT1 mutations, FLT3-ITDs, and trisomy 8. UBTF-TD AML patients had lower complete remission rates than UBTF-wild-type patients. However, their disease-free survival and overall survival did not significantly differ from those of intermediate/adverse risk patients. The study suggests that WT1-mutated clones are frequently selected under intensive chemotherapy. The study supports the recognition of UBTF-TD AML as a new AML entity in adults.
