
Vodobatinib, a selective BCR::ABL1 TKI, demonstrated significant antileukemic activity and a manageable safety profile in patients with advanced Philadelphia chromosome-positive chronic myeloid leukemia (CML) previously treated with multiple tyrosine kinase inhibitors (TKIs). In a Phase 1/2 trial, the major cytogenetic response was observed in 70% of chronic-phase patients, with significant responses also noted in accelerated- and blast-phase patients. The maximum tolerated dose was determined to be 204 mg. These findings support the need for further Phase 3 trials.
